研究室简介

Laboratory Introduction

耳鼻咽喉头颈外科研究室依托学科建设方向成立,紧密围绕临床科学问题开展一系列深入研究。现有3处办公及实验场地,分别位于成都前沿医学中心E4栋5楼、D5A栋4楼,华西临床教学楼6楼,面积1000余平。现有PI 17名,在站博士后17名,数据分析师4名,实验技术员3名,科研助理6名,以及全日制硕士、博士研究生30余人。研究室主要围绕耳鼻咽喉头颈科学开展相关研究,拥有20个特色技术平台,研究内容覆盖多组学数据分析、模式动物构建、类器官培养、基因编辑、小分子靶向递送、药物筛选、疫苗研发等多个前沿技术领域。研究室目前承担了国家自然科学基金项目(重点/面上/青年)、科技部重点研发计划、四川省科技厅重点项目和四川大学华西医院学科卓越发展1•3•5工程项目等纵向项目百余项。研究室与加拿大多伦多大学玛格丽特公主医院、西班牙加泰罗尼亚生物工程研究所、德国埃森大学、新加坡国立大学、比利时根特大学、瑞典卡罗林斯卡大学、香港大学、香港中文大学、香港城市大学等多所国际著名高校、科研院所开展紧密合作。在头颈肿瘤、耳聋遗传、内耳发育、上呼吸道免疫、类器官模型、基因编辑、疫苗免疫、高分子生物材料、喉移植等多个领域进行了一系列深入研究,相关成果发表于Nature communication、Science Advance、Elife、npj Vaccine、Science Signaling and Translational Therapeutics、Journal of Allergy and Clinical Immunology、Allergy、Journal of Clinical Immunology、Ear and Hearing、Hearing Research等杂志上。

The Otolaryngology-Head and Neck Surgery Research Laboratory was established in alignment with the department‘s academic development focus, conducting in-depth research closely tied to clinical scientific questions. The laboratory operates across three locations: the 5th floor of Building E4 and the 4th floor of Building D5A at the Chengdu Frontier Medical Center, and the 6th floor of the West China Clinical Teaching Building. These facilities cover a total area of over 1,000 square meters. The research team consists of 17 Principal Investigators (PIs), 17 postdoctoral researchers, 4 data analysts, 3 laboratory technicians, 6 research assistants, and more than 30 full-time master’s and doctoral students. The laboratory focuses on various aspects of otolaryngology-head and neck science, supported by 20 specialized technical platforms. Key research areas include multi-omics data analysis, model organism development, organoid cultivation, gene editing, small-molecule targeted delivery, drug screening, and vaccine development. Currently, the laboratory is undertaking over 100 projects, including grants from the National Natural Science Foundation of China (Key/General/Young Investigator Programs), the Ministry of Science and Technology’s Key R&D Program, the Sichuan Provincial Science and Technology Department, and the "1•3•5" Excellence Development Project at West China Hospital, Sichuan University.

The laboratory collaborates closely with prestigious international institutions such as Princess Margaret Cancer Centre (University of Toronto, Canada), the Institute for Bioengineering of Catalonia (Spain), the University of Duisburg-Essen (Germany), the National University of Singapore, Ghent University (Belgium), Karolinska Institute (Sweden), the University of Hong Kong, the Chinese University of Hong Kong, and the City University of Hong Kong. Our extensive research spans head and neck oncology, hereditary deafness, inner ear development, upper respiratory immunity, organoid models, gene editing, vaccine immunology, polymer biomaterials, and laryngeal transplantation. Research findings have been published in leading journals such as Nature Communications, Science Advances, eLife, npj Vaccines, Science Signaling, Translational Therapeutics, Journal of Allergy and Clinical Immunology, Allergy, Journal of Clinical Immunology, Ear and Hearing, and Hearing Research.


耳鼻咽喉头颈外科研究室与罕见病研究院院长袁慧军教授建立紧密合作关系,联合成立耳鼻喉-罕见病联合实验室,围绕罕见病致病基因鉴定、多组学融合分析解析疾病基因调控机制、大人群遗传变异数据库建设、罕见病靶向药物及诊治新技术开发、罕见病精准诊治智能决策支持系统开发等方面开展多项合作课题。

The Research Laboratory of Otorhinolaryngology-Head and Neck Surgery and the Director of the Institute of Rare Diseases, Professor Yuan Huijun, have established a close cooperative relationship and jointly established the Otorhinolaryngology-Rare Diseases Joint Laboratory to conduct a number of cooperative research projects on gene identification for rare diseases, multi-omics integration analysis to elucidate the gene regulatory mechanisms of the diseases, construction of a large population genetic variation database, development of targeted drugs and new diagnostic and treatment technologies for rare diseases, and development of intelligent decision support systems for precise diagnosis and treatment of rare diseases.

三、队列及样本库建设

Cohort and Biobank Development

   在科室领导、临床医疗组长、慢病随访办公室、华西生物样本库、临床研究管理部、老年医学中心、信息中心、华西生物医学大数据中心、深底医学中心的大力支持下,我科建立了头颈肿瘤慢病随访队列、老年性聋社区队列、助听干预队列,同时参与了华西医院10万例自然人队列的建设。本研究室构建了我科专病样本库及WCENT专病数据库,在场地、硬件、软件、人力方面进行长远布局,为我科战略性学科建设内容之一。数据库已实现入组患者全周期随访数据及样本智能化管理,可进行多中心、多人、多端口数据连载集成分析,集高效智能化数据采集、智能化质量控制、标准化研究设计、规范化数据分析等功能于一体,为我科临床及基础科研奠定坚实的数据及样本基础。

With the support of the department leadership, clinical medical team leaders, the chronic disease follow-up office, the Sichuan University West China Hospital Biological Sample Bank, the clinical research management department, the Center for Geriatric Medicine, the information center, the West China Biomedical Big Data Center, and the Shenbi Medical Center, our department has established a head and neck cancer chronic disease follow-up cohort, a community cohort for age-related hearing loss, and an intervention cohort for hearing aids. At the same time, our department has participated in the construction of the 100,000-case natural person cohort at West China Hospital. Our research center has built a special disease sample library and a WCENT special disease database, and has made long-term layouts in terms of venue, hardware, software, and manpower. It is one of the strategic academic construction content of our department. The database has realized the intelligent management of the full cycle of follow-up data and samples for enrolled patients, and can conduct serial and integrated analysis of multi-center, multi-person, and multi-port data. It integrates efficient and intelligent data collection, intelligent quality control, standardized research design, and standardized data analysis functions into one, laying a solid data and sample foundation for our department's clinical and basic research.

1) 大型自然人群队列数据挖掘,开展全基因组关联分析、共病遗传、环境基因交互、孟德尔随机化等研究;

2) 围绕耳聋遗传,结合进行全基因组测序数据以及家系数据进行Townes-Brocks综合征(TBS)、CD164突变、Kcnq4点突变、Pendrin致病机制相关研究;

Conduct research on the inheritance of hearing loss, combining whole-genome sequencing data and pedigree  mdata to study the Townes-Brocks Syndrome (TBS), CD164 mutations, Kcnq4 point mutations, and the pathogenic mechanisms of Pendrin.

3) 围绕内耳发育、药物性耳聋、胆脂瘤及中耳炎等开展相关机制研究。

1)大型自然人群队列数据挖掘,开展全基因组关联分析、共病遗传、环境基因交互、孟德尔随机化等研究;

A series of clinical and mechanistic studies have been conducted on HPV-related head and neck tumors (including benign and malignant tumors), focusing on immune responses and regulation, immune microenvironment, therapeutic tumor vaccines, novel tumor antigens, tumorigenesis and progression, etc. Based on multi-omics data analysis, organoid models, gene editing, and model animal construction, in-depth studies have been conducted.

Specializing in the etiology, pathogenesis, and treatment strategies of nasal mucosal inflammatory diseases, including sinusitis and nasal polyps, as well as allergic rhinitis, etc. Currently, research on the pathogenesis of sinusitis and nasal polyps is being conducted, including the role and mechanism of non-coding RNA in sinusitis and nasal polyps, as well as the role and mechanism of macrophage polarization in sinusitis and nasal polyps.
